چکیده

The structure of tubulin shows paclitaxel (Taxol(R)) binding to a pocket in beta tubulin on the microtubule's inner surface, which counteracts the effects of GTP hydrolysis occurring on the other side of the monomer.

How does taxol stabilize microtubules?

BACKGROUND The antimitotic agent taxol is an important new drug for the treatment of certain cancers. It blocks the cell cycle in its G1 or M phases by stabilizing the microtubule cytoskeleton against depolymerization. RESULTS We have used electron cryomicroscopy and image analysis to investigate the structure of microtubules assembled in vitro, and found that their fine structure was sensiti...

Reorganisation of the microtubular cytoskeleton by embryonic microtubule-associated protein 2 (MAP2c).

Microtubule-associated protein 2c (MAP2c) is one of a set of embryonic MAP forms that are expressed during neuronal differentiation in the developing nervous system. We have investigated its mode of action by expressing recombinant protein in non-neuronal cell lines using cell cDNA transfection techniques. At every level of expression, all the MAP2c was bound to cellular microtubules. At low MA...

Alterations in Ovarian Cancer Cell Adhesion Drive Taxol Resistance by Increasing Microtubule Dynamics in a FAK-dependent Manner

Chemorefractory ovarian cancer patients show extremely poor prognosis. Microtubule-stabilizing Taxol (paclitaxel) is a first-line treatment against ovarian cancer. Despite the close interplay between microtubules and cell adhesion, it remains unknown if chemoresistance alters the way cells adhere to their extracellular environment, a process critical for cancer metastasis. To investigate this, ...

Low potency of taxol at microtubule minus ends: implications for its antimitotic and therapeutic mechanism.

In many cells, low concentrations of Taxol potently block mitosis at the transition from metaphase to anaphase, with no change in microtubule polymer mass and no microtubule bundling. Mitotic block ultimately results in apoptotic cell death and appears to be the most potent antitumor mechanism of Taxol (M. A. Jordan et al., Cancer Res. 56: 816-825, 1996). Mitotic inhibition results, at least in...

Taxol-induced bundling of brain-derived microtubules

Taxol has two obvious effects in cells. It stabilizes microtubules and it induces microtubule bundling. We have duplicated the microtubule-bundling effect of taxol in vitro and report preliminary characterization of this bundling using electron microscopy, sedimentation, and electrophoretic analyses. Taxol-bundled microtubules from rat brain crude extracts were seen as massive bundles by electr...
